What platform do you play on? PC players using the unofficial patches will probably not be able to get it to work, as it relies on "Restoration looping" (i.e., using one Restoration potion to augment a second Restoration potion). This is an exploit that the unofficial patches have fixed.
If you're not on PC (or if you're just not using the unofficial patches) then I'm not sure. The only thing I can think of is that you're not following the instructions correctly. Also be aware that, while we said the trick can work with other items besides the Locket of St. Jiub, this particular combination of potions does require you to be using an item with +50 Fortify Stamina -- no more, no less. So if you're trying to do it with an item other than the locket, make sure it has +50 Stamina.
It also may not work if you've got any other items/effects that modify Stamina -- this could include enchantments, blessings, diseases, level-up attribute allocation, etc. Check your "Active Effects" tab in the Magic menu to make sure there are no other effects modifying your character's Stamina.

Yup, that'll do it. You can use the Gauldur Amulet to get infinite Stamina, you just need to use a different potion combination: First, drink a Potion of the Healer; then drink a Philter of the Healer; then equip the Gauldur Amulet. That should do it!

At Alchemy 20 with no perks or enchantments, make a few potions using a combination of any two of Abecean longfin, Cyrodilic spadetail, and salt pile. You can get these fishies in Lake Ilinalta or fish barrels across Skyrim. You should get a magnitude of 18% Fortify Resto on them. Now, grab the first Alchemist perk and make some 21% magnitude potions. Drink the 18%, then the (formerly) 21%, and equip any +30 stamina item to get the infinite stamina glitch! This one's nice because you don't have to wait until level 10 or punch merchants to get the right premade potions.

As usual with builds Teccam is involved in, getting an estate with a nice greenhouse is recommended. This build doesn't use tons of fishies, but Windstad is still helpful for those river betties!
